Cytopathic effect of coccoid forms of Helicobacter pylori in Albino rats and Swiss mice

چکیده
The effect of coccoid forms of a vaculating cytotoxin A positive Helicobacter pylori strain on Mus musculus (Swiss mice) and Rattus norvegicus (Albino rats) was studied. Two groups of Swiss mice and Albino rats were used. The first group of mice and rats was orally inoculated with coccoid forms of Helicobacter pylori, whereas, the second group of mice and rats were untreated and used as a control. The animals that had been orally inoculated with a total count of 7.0x10 bacterial cell / ml died after 5-7 days of inoculation (mice), whereas, rats died after 7-9 days. Symptoms of mouth ulcers, darkening of the ventral part and peripheries and loss of weight were apparent, in addition to slow movement and general weakness compared to the control group. Histological examination of the stomach of the inoculated animals showed marked degeneration of the epithelial lining membrane and dark dense bodies most probably coccoid forms inside gastric glands. In addition dialated blood vessel, degenerated, vacuolated fused cells with coccoid forms and intravascular haemolysis were observed. These findings may indicate the toxicity of coccoid forms of Helicobacter pylori and hence their possible pathogenic role in humans. [Rajaa M. Milyani. Coccoid forms of Helicobacter pylori.
To the Editor-Eaton et al. [1] recently described experiments with coccoid Helicobacter pylori in gnotobiotic piglets. This interesting study examined the colonization and the reversion from coccoid to bacillary H pylori within 2 weeks of inoculation. Piglets colonized with bacillary H pylori had more severe gastritis 2 weeks after inoculation than did those sacrificed 1 week after inoculation....
متن کاملThe Immunohistochemistry and Toluidine Blue Roles for Helicobacter pylori Detection in Patients with Gastritis
Background: Helicobacter pylori, which is associated with many upper gastrointestinal diseases, is found in half of the population of the world. Several special stains and immunohistochemistry stain for H. pylori are available. The need for and usefulness of immunohistochemical (IHC) technique has been debated for years. Toluidine blue is a simple stain for microbiological studies and is easily...
متن کاملCoccoid Helicobacter pylori Can Directly Adhere and Invade in Agminated Formation to Human Gastric Epithelial Cells
Helicobacter pylori (H. pylori) can infect into the epithelial cell to cause benign or malignant disorders. Under stressful environment, a spiral form of H. pylori is transformed into a coccoid form. The infectivity of the coccoid form is still controversial. Since spiral forms are transformed into two types of coccoid forms via different mechanisms, the infectivity of the two types of coccoid ...
متن کاملMorphological and Bactericidal Effects of Different Antibiotics on Helicobacter pylori
BACKGROUND Helicobacter pylori (H. pylori) is a spiral Gram negative bacteria that can transform to the coccoid form in adverse conditions. OBJECTIVES The aim of this study was to determine the in vitro morphological and bactericidal effects of metronidazole, amoxicillin and clarithromycin on H. pylori. MATERIALS AND METHODS The standard strain 26695 of H. pylori was cultured on Brucella ag...
متن کاملCoccoid forms of Helicobacter pylori are the morphologic manifestation of cell death.
Helicobacter pylori can transform from its normal helical bacillary morphology to a coccoid morphology. Since this coccoid form cannot be cultured in vitro, it has been speculated that it is a dormant form potentially involved in the transmission of H. pylori and in a patient's relapse after antibiotic therapy. In this study we determined the effects of aging, temperature, aerobiosis, starvatio...
